I was intrigued by the mechanisms of immunity, especially generation of diversity (GOD), that protect and at the same time are tolerant to self constituents. The loss of tolerance, however, leads to development of autoimmune disease occasionally. While in France, United States and Switzerland, our research on construction and regulation of immune system in health and disease demonstrated that the same genetic elements of the immune system, some identified by us earlier, defended a host against harmful substances or agents and also led to the origin of pathogenic autoantibodies responsible for systemic autoimmune disease. Our laboratory also demonstrated that B-cell selection early in the development influenced the generation of pathogenic autoantibodies. Such a school of thinking is outlined the book ‘Molecular Immunobiology of Self-Reactivity’ published in 1992 by Marcel Dekker Inc., New York, USA.

   

Studies of the construction of immune system across species led us to focus on the bovine immune system where we identified, classified the genetic elements encoding bovine antibody and defined the mechanisms of antibody diversification. We discovered that limited germline diversity in the cattle genome necessitated the origin  of unique largest sized antibody, yet to be seen in any other species, where an exceptionally long CDR3H is generated resulting in novel configurational diversity. Our current research focuses on structural and functional aspects of antibody engineering for developing next generation of  immunodiagnostics, immunotherapeutics (e.g., construction of single chain Fv (scFv) and novel vaccines.
