Agilent TapeStation 4150 Service
The TapeStation system allows for automated electrophoresis of most DNA and RNA samples. Typical applications include quality control (QC) of NGS libraries and RNA extractions (ie. RIN). This method excels in the determination of fragment length distribution of nucleic acid samples, but is not typically relied on for accurate quantification. The TapeStation can be used in place of the BioAnalyzer for most applications, but cannot currently be used for small/micro RNA analysis.
Service includes Standard and High Sensitivity assays for RNA, DNA >1kb, and DNA >5kb, as well as a Standard gDNA assay.

DNA Sequencing
Sample submission considerations.
The facility provides a number of standard sequencing primers for common plasmid vectors.
Custom primers should be provided at a concentration of 5 uM (=5 pmol/uL).

Next Generation Sequencing
The Illumina NextSeq 1000 is an NGS sequencing system designed to miniaturize sample and reagent volumes and increase read output. The NextSeq 1000 is capable of many sequencing applications, including targeted metagenomics, whole genome sequencing, transcriptomics, immune repertoire profiling, sequencing of targeted panels, exomes and large panels, circulating tumor DNA, single-cell sequencing, and spatial profiling.

Nucleic Acid Fluorometry (Qubit)
Fluorometric concentration assays are generally more accurate than spectrophotometric/UV-Vis absorbance (ie. Nandrop) measurements because they are less affected by common contaminants (ie. Proteins, RNA, or DNA).
This method is recommended NGS library assessments and for any application that requires higher accuracy in concentration estimates than can be acheived with Nanodrop.

PromethION Services
Plasmid & PCR Amplicon Sequencing
The PromethION 24 is a high-throughput nanopore sequencer capable of natively sequencing both RNA and DNA with rapid turn-around times. The PromethION can load up to 24 flow cells, each capable of multiplexing 96 samples or more. It is also possible to re-use flow-cells, decreasing cost and waste. The PromethION 24 is suitable for long-read applications, including genome assembly, native methylation seq., direct RNA transcript sequencing (including alternate splicing), and metagenomics.
We have phased out Sanger Sequencing in favor of long-read sequencing on the Oxford Nanopore PromethION 24. The facility provides sequencing of DNA fragments up to 25 kilobases in length or more, including whole plasmids and PCR amplicons. Service starts on September 2.

Sequencing Request Form - Use this form to submit DNA templates (e.g. PCR amplicons, Plasmids) for sequencing. Recommended at least 100 ng DNA. Recommended concentration 10 ng/µL in 10 µL in molecular grade water or TE buffer. Concentration should be estimated by fluorometric methods such as Qubit/PicoGreen.
- Fill out all three tabs of the request form (first two tabs are mandatory)
- Signed request form emailed to genomics@uoguelph.ca
- Drop the samples off at room SSC 1401 with facility staff or in the Sample Drop-Off Freezer
